I have recently done very similar to one side (yet to to the other) of my GF's supra.
I installed Peerless HDS phase lug 6inch mids though.
First was to strip the door down, ie door card, undo the power window motor, and some of the arm rest brackets so they are out of the way. Remove the plastic sheet also.
Then cleaned inside the door skin (outer) with presol and applied Dynamat Exteme panels to all I could get to.
After this I then made templates to fill the two larger holes, one towards the rear of the car and the one that the power window motor/cables feed into also. Both from 6mm MDF
Then I set about the large/front speaker panel. Using 6mm MDF I made a panel to cover and 'seal' as much of the larger opening as seen in your pictures. I had a couple of 6.5inch mid rings made from 18mm MDF that I fastened to this panel with a single screw for starters, and trial fitted the card back over the door to make sure the MDF ring was centred on the speaker hole in the door card. Once happy I screwed and glued it through the back of the 6mm MDF and cut out the inner shape of the ring from the 6mm MDF to create a cavity.
I then followed the tute on this forum about sound deadening doors and layed a matrix of duct tape over the holes totally sealing them(or as best I could) before inserting the MDF panels and again sealing over the top of them with Duct tape. Once happy with the seal I put more dynamat extreme over the whole inner door panel and sealed the newly placed panels with the deadener sheets.
Once I was happy I had most of the door sealed I ran some speakers wires to the speaker and poked them out the speaker hole, put some closed cell foam sealing tape on the MDF speaker ring and then put the door back together and fitted the door card.
Once all together I put the speaker OVER the door card and actually clamped the door card air-tightly to the closed cell foamed MDF ring with screws. This looks very neat and tidy, and once I change the screws for black ones will be a very smooth looking install. I am not using grills for the moment, but will look into making some custom 'rod' grills at a later date to stop VEY stray kness peircing themselves on the phase lugs
